A typical word order in Japanese is "the subject, the object and the verb."
O is a particle indicating the object of an action.
e.g.) WATASHI WA MANGA O YOMIMASU. (I read manga.)

How to make a verb into a negative and a question
We call the verbs that end with MASU "the MASU-form of verbs." We use the MASU-form, when we speak politely. To make it a negative, we change MASU to MASEN. So, YOMIMASU (to read) becomes YOMIMASEN.
